Lyse cells 4 hours to overnight in 500ul Lysing Solution (with proteinase K and 2- mercaptoethanol). Incubate at 55 ordm;C.

Add fresh to aliquots of solution:

5ul 2-mercaptoethanol (not included) per 1ml lysing solution

20ul Proteinase K (20mg/ml) per 1ml lysing solution

2.Vortex 10 seconds. Add 250ul Salt Solution and vortex 5 seconds. Let stand 15 minutes on ice.

3.Spin 15 min at maximum speed at 4 ordm;C

4.Transfer supernatant to clean tube and precipitate DNA with 750ul ethanol. Mix well. Pellet by spinning gently at room temperature at speed “10” for 2-3 minutes.

5.Wash DNA pellet with 750ul 70% ethanol and air dry. Dissolve in 100ul TE or DNase-free water. Resuspend DNA rapidly by incubating 15 minutes at 55 ordm;C
